A streamlined 2.5-hours workflow:
Performance:
1. Minimal GC bias similar to mechanical shearing: WGS Fragmentation Mix shows similar coverage distribution to Covaris mechanical shearing and cover across extreme GC% spectra at 1ng-100ng DNA input, and it performs the best among similar products.
Figure 1: 3 bacterial genomes with different GC content equal molar mix were conducted library preparation based on the recommend protocol.
2. Flexibly adjustable fragment sizes: The desired fragment size can be achieved by adjusting the shearing time.
Figure 2: 1μg of input DNA were fragmented using the fragmentation mix for different times that 3 minutes, 6 minutes, 10 minutes, 12 minutes, 15 minutes, and 18 minutes. Data were generated and analyzed using an Agilent® Bioanalyzer 2100 High sensitivity Chip.
3. Superior DNA library yield: The library yield is significantly higher than that of competitors before or after amplification.
Figure 3: The library constructions were use by different brands of library preparation kits. The library concentration was evaluated prior to amplification.
Figure 4: After DNA fragmentation by Covaris and library construction, samples with different input amounts were amplified using different brands of kits and compared for library yield. The amplification cycle numbers were kept consistent, and library quantification was performed using Qubit fluorescence quantification and qPCR.
4. High reproducibility & consistency: Superior reproducibility and consistency between run-to-run, genome-to-genome and lot-to-lot.
Figure 5: Very consistent fragmented DNA was generated across 4 replicates using the same input DNA and the same lot of the WGS fragmentation mix using fragmentation time 5min and 10min.
Figure 6: Very consistent fragmented DNA was generated across the 2 different lots of the WGS fragmentation mix on the same samples using fragmentation time 5min and 10min.
