Tumor-selective pro-immunocytokine (IFN-ICB) platform that links interferon (IFN) to an immune checkpoint blockade (ICB) via a tumor-specific metalloproteinase (MM)P-cleavable substrate. Both components remain inactive in normal tissues and are reactivated in tumors, enhancing MHC expression and T cell activity. In a murine colorectal cancer model, IFN-ICB improved efficacy by 50% and safety by 75%. This technology also successfully masked multiple ICB antibodies, validating the platform’s versatility. The dual-shielding strategy offers a safer, versatile alternative to conventional immunocytokines.
