Objectives:
Develop a highly stable and high-penetration NMN delivery system, prepare NMN-loaded liposomes through a Taylor reactor (TCR), and solve the problems of rapid decomposition and poor absorption of NMN in traditional drug delivery methods.
Research Background:
NMN is a precursor of NAD⁺ and has the potential to fight aging, metabolic disorders and neurodegenerative diseases. However, oral NMN is easily metabolized into NAM in the gastrointestinal tract, which greatly reduces its effectiveness. Transdermal delivery via liposomes can bypass gastrointestinal metabolism and enhance in vivo absorption and efficacy, but traditional methods for preparing liposomes (such as thin film hydration method) are often limited by the use of organic solvents, uneven particle size, and poor encapsulation efficiency.
Research Methodology:
Liposomes were prepared using TCR, a system that allows precise control of shear forces and fluid mixing, helping to form liposomes with uniform particle size. The study designed experiments with different rotation rates (rpm) to analyze their effects on particle size, Zeta potential, stability and penetration effect, and conducted ex vivo porcine skin penetration tests (Franz cell diffusion method) and HPLC quantitative analysis to evaluate the performance of liposomes.
Study Features:
** First use of TCR to prepare NMN liposomes
** Liposome particle size can be stably controlled in the range of 125–145 nm
** Zeta potential reaches -40 mV, showing good stability
** After 180 days of storage at room temperature, more than 87% NMN activity is still retained
** Penetration of pig skin reached 96.8% within 12 hours, significantly better than traditional microneedle therapy
Research results:
The TCR preparation method is not only simple to operate and does not require organic solvents, but can also prepare liposomes with high stability, high encapsulation efficiency, and uniform particle size in a short period of time, showing its potential as a future NMN delivery platform. Especially in the fields of anti-aging, beauty and biomedicine, it has high application value.
